The adsorption of procyanidins (condensed tannins) on cell-wall material was quantified by bringing into contact solutions of procyanidins and suspensions of cell-wall material. A model was developed on the basis of the Langmuir isotherm formulation and a factorial experimental design. The parameters that influenced the adsorption were the concentration and molecular weight of the procyanidins, the ionic strength of the solution, the temperature, and the apple cell-wall concentration. The model was applied to partitioning of procyanidins from apple between juice and mash. The parameters to be taken into account are the composition of the apples and, specifically, (i) the concentration and molecular weight of the procyanidins, (ii) their acidity and pH as a determinant of the ionic strength, and (iii) their cell-wall content and the temperature at pressing. To estimate the ability of the model to relate procyanidin concentrations in the juice to their concentration in the apple, apples of three varieties of widely different procyanidin compositions were pressed in conditions that prevent oxidation. In these conditions, yields in the juice were >80% for phenolic acids or catechin monomers but <50% for procyanidins, with the lowest rates obtained for the higher polymers in accordance with the model.  相似文献

Phenolics from grapes and wines can play a role against oxidation and development of atherosclerosis. Stilbenes have been shown to protect lipoproteins from oxidative damage and to have cancer chemopreventive activity. We describe a method for the direct determination of stilbenes in several red wines using high-performance liquid chromatography with UV detection. In a survey of 12 commercial wines from the south of Brazil (Rio Grande del Sul), levels of delta-viniferin are reported for the first time in different varieties of red wines. Brazilian red wine contains trans-astringin, trans-piceid, trans-resveratrol, cis-resveratrol (in high quantity: 5 times more than the trans form), epsilon-viniferin, and a compound isolated for the first time in wine, trans-delta-viniferin. Isolation and identification of delta-viniferin was achieved by NMR after extraction and fractionation of red wine phenolics. delta-Viniferin contributes, as well as cis-resveratrol and trans-piceid, to a significant proportion of stilbenes in wine dietary intake, particularly with Merlot varieties containing an average level of 10 mg/L for delta-viniferin, 15 mg/L for cis-resveratrol, and 13 mg/L for trans-piceid. The total stilbene intake from wine origin was estimated for the Brazilian population as 5.3 mg/day per person (on the basis of a regular wine consumption of 160 mL/day). delta-Viniferin can contribute to around 20% of total stilbenes in wine (average of 6.4 mg/L in red Brazilian wines). It would be important in the future to investigate the origins of the differences in wine stilbene levels in relation to the vine varieties, and the bioavailability of the newly extracted stilbene delta-viniferin in plasma after consumption of different types of wines.  相似文献

Rumelenic (cis-9,trans-11,cis-15 18:3) acid is a naturally occurring conjugated isomer of alpha-linolenic acid (CLnA) in milk fat. Metabolism in rats was studied using a synthetic CLnA mixture, composed mainly by equimolar quantities of cis-9,trans-11,cis-15 and cis-9,trans-13,cis-15 CLnA isomers. Their metabolisms were studied by feeding high quantities of CLnA (150 mg/day) for 4 days to rats that had been reared on a fatfree diet for 2 weeks. After this period, animals were sacrificed and liver and epididymal adipose tissue lipids extracted. Six metabolites of the cis-9,trans-11,cis-15 18:3 CLnA isomers were identified as being cis-7,trans-9,cis-13 16:3, cis-11,trans-13,cis-17 20:3, cis-8,cis-11,trans-13,cis-17 20:4, cis-5,cis-8,cis-11,trans-13,cis-17 20:5, cis-7,cis-10,cis-13,trans-15,cis-19 22:5, and cis-4,cis-7,cis-10,cis-13,trans-15,cis-19 22:6 acids. Two metabolites of cis-9,trans-13,cis-15 18:3 CLnA isomer were also identified by GC-MS as being cis-7,trans-11,cis-13 16:3 and cis-5,cis-8,cis-11,trans-15,cis-17 20:5.  相似文献

In European forests, plants, fungi and invertebrates have been proposed as indicator species for assessing conservation value at the stand scale. To cover larger spatial scales, wide-ranging vertebrates could be added to that set of species. For resident forest birds, we (1) explored whether the occurrence of some species could be used to indicate high species richness and abundances, and (2) compared the results among four regions in the Baltic Sea drainage basin with a common species pool but different forest management intensities and varying proportions of deciduous and coniferous trees (south-central and southern Sweden, south-central Lithuania and northeastern Poland). Assemblages of deciduous forest birds in 100-ha landscape units were generally nested, suggesting that species richness within that group may be predicted based on the presence of a few species. Birds of coniferous forests, however, showed poorer conformity to nestedness in Sweden. Specialised species such as the middle spotted (Dendrocopos medius) and lesser spotted woodpeckers (D. minor) in deciduous forest and the three-toed woodpecker (Picoides tridactylus) in coniferous forest generally figured among the best indicators. In deciduous forest, there was high cross-regional consistency in the identity of the best indicators. Moreover, the sites where the best indicator was present also harboured higher relative abundances of most background species. For coniferous forest, however, such a relationship was not found. We conclude that an indicator species approach may be useful for resident birds of deciduous forests in hemiboreal Europe, emphasising that it should constitute one of many complementary tools for conservation management.  相似文献

The occurrence of biologically active compounds identified for the first time in the lipophilic extracts of yellow birch (Betula alleghaniensis Britt.) foliage led to the quantification of the seasonal variation of their concentrations. Yellow birch foliage was collected from late June until late September 2003 in two different regions of Quebec. The extraction yields using hexane as a solvent were determined, and the extracts were analyzed by GC-MS to identify their molecular composition. In terms of both extraction yields and the concentration of the targeted molecules present in the extracts, mid-September has been determined as the best time to collect foliage samples. A total of 14 constituents were identified in these extracts. This is the first report of the presence of all of these constituents in yellow birch foliage and of some of them in the genus Betula. The most important compounds identified in yellow birch foliage extracts are triterpene squalene and aliphatic hydrocarbon tetracosan, aliphatic alcohol phytol, fatty acids hexadecanoic and octadecanoic, pentacyclic triterpenes alpha- and beta-amyrin, and phytosterol stigmast-5-en-3-ol.  相似文献

Stilbenes have been shown to have cancer chemopreventive activity and to protect lipoproteins from oxidative damage. A method for the direct determination of stilbene oligomers (viniferin and pallidol) as well as astilbin in different types of wine using high-performance liquid chromatography with UV detection is described. In a survey of 21 commercial wines from the south of France, levels of pallidol and viniferin are reported for the first time in different types of wines. Viniferin was found to be present only in red and botrytized sweet white wines with levels between 0.1 and 1.63 mg/L; pallidol was not found in dry and sweet white wines but only in wines made by maceration with stems, with levels between 0.38 and 2.22 mg/L. Highest levels of astilbin were found in Egiodola (15.13 mg/L), Merlot (11.61 mg/L), and Cabernet Sauvignon (8.24 mg/L) for red wines and in Sauvignon (5.04 mg/L) for white varietal wines. Astilbin levels are highest for recent vintages, but pallidol is not found in older vintages. During noble rot development in Sauvignon or Sémillon grapes from the Sauternes area, levels of trans-astringin, trans-resveratrol, trans-piceid, and pallidol are quite low (<0.5 mg/kg of grapes). Viniferin and astilbin levels become optimum at 2 and 30 mg/kg, respectively, during spot grape and speckle grape stages.  相似文献

Potential oxygen consumption by lees, more precisely by nonviable yeasts, during wine aging was recently described. Additionally, yeast autolysis is described as the main mechanism of degradation of lees during wine aging. Thus, to understand the effect of oxygen consumption by yeast lees during wine aging, an accelerated wine aging methodology was tested. Wine aging in the presence of yeast lees was studied both in the presence and in the absence of oxygen. Different markers of yeast autolysis were followed to find a relationship between oxygen consumption by yeast lees and changes in the final wine composition after aging. No differences for compounds tested were found in the wine and in the lees except among sterol compounds in lees: in the presence of oxygen, the concentration of ergosterol in lees was significantly lower than that in the absence of oxygen. It was hypothesized that ergosterol could be oxidized under the influence of oxygen, but none of the known products of ergosterol oxidation were recovered in the corresponding yeast lees. In addition, the decrease of ergosterol content in yeast lees cannot account for the total amount of oxygen consumed by yeast lees during such wine aging.  相似文献

ABSTRACT: Staphylococcus aureus is a major cause of mastitis in ruminants. In ewe mastitis, symptoms range from subclinical to gangrenous mastitis. S. aureus factors or host-factors contributing to the different outcomes are not completely elucidated. In this study, experimental mastitis was induced on primiparous ewes using two S. aureus strains, isolated from gangrenous (strain O11) or subclinical (strain O46) mastitis. Strains induced drastically distinct clinical symptoms when tested in ewe and mice experimental mastitis. Notably, they reproduced mild (O46) or severe (O11) mastitis in ewes. Ewe sera were used to identify staphylococcal immunoreactive proteins commonly or differentially produced during infections of variable severity and to define core and accessory seroproteomes. Such SERological Proteome Analysis (SERPA) allowed the identification of 89 immunoreactive proteins, of which only 52 (58.4%) were previously identified as immunogenic proteins in other staphylococcal infections. Among the 89 proteins identified, 74 appear to constitute the core seroproteome. Among the 15 remaining proteins defining the accessory seroproteome, 12 were specific for strain O11, 3 were specific for O46. Distribution of one protein specific for each mastitis severity was investigated in ten other strains isolated from subclinical or clinical mastitis. We report here for the first time the identification of staphylococcal immunogenic proteins common or specific to S. aureus strains responsible for mild or severe mastitis. These findings open avenues in S. aureus mastitis studies as some of these proteins, expressed in vivo, are likely to account for the success of S. aureus as a pathogen of the ruminant mammary gland.  相似文献
